Ontogeny of mRNAs for gpd-4 and gpd-2. 10 g of total embryonic (E) and postembryonic larval (L4) mRNAs were dot blotted on to nitrocellulose paper and probed with gpd-2 and gpd-4-specific probes. The probe for gpd-2 was prepared using pSS1.3 (Fig. 3) derived from an M13 phage DNA using an oligonucleotide primer located 196 bp from its termination codon, which extended to a HaeIII site 5 bp before its stop codon. Since the 5' end of gpd-3 obtains a trans-spliced leader sequence (see Results and Discussion) this probe is specific for gpd-2 only. The gene-specific probe for gpd-4 was prepared using pB9 (Fig. 1(b)), an oligonucleotide 164 bp downstream from its termination codon, which was then extended to a HaeIII site located 10 bp beyond its stop codon. | |||
