Subcellular_localizationIn embryo, CeVPS-32 immunoreactivity was detected in a diffuse cytosolic pattern with a punctiform component characteristic of vesicular localisation. To confirm that CeVPS-32 positive vesicles are endosomes authors looked for colocalization with the endosomal marker CeVPS-27/HGRS-1. Vesicles positive for both CeVPS-32 and CeVPS-27 were detected in all epithelial tissues. However, only a subset (30%, n=606) of vesicles is positive for both markers suggesting that the presence of CeVPS-27 and/or CeVPS-32 correspond to distinct stages of endosomal maturation.

The specificity of anti-CeVPS-32 antibodies was confirmed by the almost complete extinction of the signal after inactivation of Cevps-32 by RNA interference and in the Cevps-32(ok1355) null mutant.
