PatternPgpdh-1::GFP expression was virtually undetectable under control conditions but showed dramatic up-regulation in the intestine and hypodermis in worms exposed to hypertonicity.
RemarkPgpdh-1-GFP expression was proportional to the level of hypertonic stress, and the time course of expression correlated well with that of glycerol accumulation. Heat shock (30 centigrades), endoplasmic reticulum stress (10 ug/ml tunicamycin), cold (4 centigrades), and oxidative stress (230 uM juglone and 100% O2) failed to activate Pgpdh-1::GFP. These data demonstrate that Pgpdh-1::GFP specifically reports activation of signaling pathways required for glycerol synthesis and adaptation of C. elegans to hypertonic stress.
