PatternWe first used a previously characterized dmd-3::YFP transcriptional reporter that contains 4264 nucleotides upstream of the start codon, the first exon, the first intron and part of the second exon fused to YFP (Mason et al., 2008) to examine the dynamics of dmd-3 expression in the linker cell and hindgut in L1 - L4 males. We found that dmd-3: YFP expression initiates in the linker cell in L3 males when the linker cell is midway through stage II and persists until linker cell engulfment and death. In addition, we observed that dmd-3::YFP is expressed in a subset of posterior presumptive vas deferens cells that contact the trailing side of the migrating linker cell. Expression in the vas deferens initiates during or closely following the turning of the linker cell to the ventral side (mid-stage III) and persists as the linker cell migrates to the hindgut (stage IV). dmd-3::YFP expression in the hindgut cells of the U lineage commences in L1/L2 males, coincident with stage I of linker cell migration, and persists through linker cell engulfment and hindgut morphogenesis in late L4 males. We do not observe dmd-3::YFP expression in the distal tip or hindgut cells of L4 hermaphrodites (Mason et al., 2008).
