Construction_summary[nep-1::gfp] transcriptional fusion constructs. To analyse the expression pattern of nep-1, 1.2 kb and 4.0 kb of promoter were amplified by PCR, using the sense primers 5$(B!l(B-CCCAAGCTTGTGTACACCTCTTGTACGTCC-3$(B!l(B and 5$(B!l(B-GACCTGCAGACAGTTTTCATGTCGGCAATCA-3$(B!l(B, respectively and a common antisense primer (5$(B!l(B-CCCAAGCTTCTTACAGGTGTAGTAAGTAGGC-3$(B!l(B). The PCR-products and the vector pPD95.75 were digested with HindIII/PstI and ligated. The resulting plasmid (the GFP sequence under the control of the nep-1 promoter) was microinjected into young adult N2 wild-type C. elegans. Both nep-1 promoter constructs induced a strong green fluorescent protein (GFP) signal, suggesting that all essential regulatory elements are located within 1.2 kb upstream of the predicted start codon. --precise ends.
