Gamma tubulin is a critical component of the microtubule organizing center (MTOC), and mediates cell division cycle in higher organisms.In C.elegans gamma tubulin is highly divergent and encoded by a single gene tbg-1,loca- ted on LGIII. Temporal expression of tbg-1 during C. elegans embryonic and post embryonic development directly correlates with cell division cycles. Expression of tbg-1 antisense RNA under the heat shock promoter results in embryonic arrest at the one cell stage reminiscent of the ts emb-30 (g53) mutant. Transformation of emb-30 mutant with a 8 kb HindIII-XhoI genomic fragment that contains the tbg-1 gene rescues the mutant phenotype. Sequence ana- lysis of the 8 kb rescuing fragment reveals a polycistor nic unit comprising 3 genes uce-1 (ubiquitin conjugating enzyme), rsp-1 (RNA polymerase 16 kd subunit), and tbg-1 (gamma tubulin), with a common promoter located 5' upstr- eam of the uce-1 gene, that are cotranscribed. The uce-1 and rsp-1 genes are trans-spliced to both SL1 and SL2 leaders; but the tbg-1 only to the SL2 leader, typical for the operon.
