Some mutations show dauer-constitutive (Daf-c) phenotypes only when two of them are combined to make double mutants*. We have made an extensive search of such synthetic Daf-c phenotypes among known mutations and found that there are more than 40 genes for them. The genes include those required for normal chemotaxis (che-2, osm-1, daf-6, tax-2, odr-1 and at least 18 other genes), locomotion (unc-3, unc-31, unc-101, unc-104), cell lineage (lin-32, vab-3), fluoride-sensitivity (flr-1 to flr-5), egg-laying (egl-4, egl-32), defecation (aex-3), catecholamine distribution (cat-1), mechanosensation (mec-1) and body size (sma-2 etc.). We think those mutations affect sensory neurons for food and temperature and/or interneurons connecting to them. Some of them can be replaced by a serotonergic antagonist ketanserine to obtain the same effect. We have also studied enhancement of the Daf-c phenotype of known daf mutations by some of those mutations, and suppression of the synthetic Daf-c phenotype by known daf (Daf-d) mutations. Finally, we classified those mutations and made models of the dauer signal processing pathway, using the pattern of combinations that cause synthetic Daf-c phenotypes.
