Under conditions of starvation and overcrowding, C. elegans may arrest development at the second molt to form a dauer larva. Dauer larvae, with specialized structure and behavior, are adapted for dispersal and long-term survival. Mutations in more than thirty daf genes have been isolated. To look for new mutations affecting dauer morphogenesis, we conducted a screen for mutants that form dauer and dauer-like larvae constitutively. Fifty-four dauer-like mutants defining at least 15 genes were isolated. These dauer-like mutants fail to respond appropriately to environmental cues, and are incapable of executing either complete dauer or nondauer development (Jia et al. Development, 129:221-231). Dauer-like larvae show some, but not all, of the normal dauer characteristics. These mutants have a high density of intestinal and hypodermal fat granules. However, the dauer-like phenotypes are not temperature sensitive, and the mutants are not SDS resistant, which are typical characteristics of constitutively formed dauer larvae. Most dauer-like mutants function either downstream or in parallel with daf-12, the main output of TGF- signaling in dauer formation. We cloned daf-31, one of dauer-like mutant genes, by the positional cloning method. The daf-31 gene encodes a putative GNAT family acetyltransferase. Biological functions of DAF-31 are being characterized.
