Figure 10. RAB-6.2 and LIN-10 colocalize with RME-8 in neurons. (A-D) Fluorescence from neuron cell bodies (A and C) and ventral cord dendrites (B and D) is shown. (A-B'') Cerulean::RAB-6.2 (A and B) colocalizes with Venus::RME-8 (A' and B'; A'' and B'' show merged). (C-D'') LIN-10::CFP (C and D) colocalizes with Venus::RME-8 (C' and D'; C'' and D'' show merged). Arrowheads indicate colabeled puncta. Bars, 5 um.
(A, A'-D, D') Expression of the frizzled homolog T23D8.1 monitored using GFP. PT23D8.1::GFP is expressed in a few cells of the nerve ring (A, A'). Further, it is expressed in the intestine (B, B'), one cell on the ventral side of the terminal bulb of the pharynx (arrow in C, C') and two cells around the anus (D, D').
(C, C') Pgrl-4::grl-4::GFP is weakly expressed in the hypodermis. Tail region is shown (arrows point to the nuclei of the hypodermal cells). (D, D') The GFP-tagged GRL-4 protein is not associated with the shed cuticle (the arrow points to the anus). A'-D' are DIC images corresponding to the fluorescent images A-D, respectively.
(D-F) Wild-type anaphase one-cell stage embryos stained with anti-tubulin (D) and anti-ZYG-8 (F) antibodies. (D)-(F) are at the same magnification. Bar = 10 um. Anti-ZYG-8 antibodies mark the spindle and spindle poles (arrowheads) in wild-type.
(A-D) Worms (wild type [A-C]; itsn-1(ok268) [D]) were stained with anti-ITSN-1 Ab (red). Worms are in lateral view, head to the left. NR, nerve ring (blue line in D); VC, ventral cord; DC, dorsal cord. Bar, 10 um (all panels).
