In case of the pan-neuronal promoters, expression under cmk-1 promoter is observed in most, if not all neuronal nuclei, including in many head neurons (C top) and ventral nerve cord (VNC) neurons (C bottom).

Reporter gene constructs were generated for genes from the SAGE library as shown. Green boxes indicate gfp coding sequences (rfp in the case of dop-3 and yfp in the case of hlh-10). If extrachromosomal arrays were generated, multiple lines were analyzed for each reporter construct, as indicated under each panel of micrographs. Each dataset of micrographs shows expression of the reporter construct under investigation in a high-magnification (400x) image, the overlap with a fluorescent marker for ASEL/ASER (in all except one case, the otIs151 transgene) and a low magnification (160x) overview of expression of the reporter construct under investigation throughout the whole worm. FIG. 2. Expression pattern of SPL GFP reporters. Transformation of worms was performed by microinjection with plasmid DNA as described under "Experimental Procedures." Several stable lines were generated for each construct and visualized by mounting live transgenic nematodes on 5% agarose and viewing by Nomarski optics and by fluorescence microscopy with a Chroma High Q GFP long pass filter. Transgenic worms containing constructs of 200, 250, and 275 nucleotides upstream of the start codon demonstrated background fluores- cence similar to the 129-nucleotide construct and are not shown. In each case >20 worms were analyzed.

In case of the pan-neuronal promoters, expression under unc-119 promoter is observed in most, if not all neuronal nuclei, including in many head neurons (D top) and ventral nerve cord (VNC) neurons (D bottom). However, in a subpopulation of animals expressing BFP under the unc-119 promoter, intense fluorescence was observed in several additional larger nuclei along the lateral sides (D, blue arrowheads), which we identify as hypodermal cells and faint signals in even larger nuclei (D, pink arrowheads), which we identify as intestinal cells.

Dynamin was also detected in sublateral neurons (sn). The inset shows that dynamin accumulates along the apical surface of intestinal cells (arrowhead). In this case, the worm was broken during the permeabilization procedure, which helped unmask the specific intestinal staining.
