Embryos costained with anti-EGG-3 and anti-EGG-4/5 antibodies. EGG-3 and EGG-4/5 relocalize from the cortex to subcortical speckles during the transition from meiosis I to meiosis II. In the 2-cell stage, occasional EGG-3-positive speckles can still be detected. By this stage, the EGG-4 signal is at background levels.

Supplemental Figure 7: MRE-11 localization is not affected in ubc-9(tm2610) mutants. GFPwas visualized by fixing worms without antibody addition. MRE-11 localizes to the nucleusthroughout prophase I. This localization does not change in ubc-9(tm2610) mutants. Polarizednuclei can still be seen in mid/late pachytene of ubc-9 germlines, which is normal for thisgenotype.

The time that isolated worms spent moving forward, backward, or still was measured. No statistically significant difference was detected between the sexes in time spent moving forward (p > 0.05) or moving backward (p > 0.05). Males, on average, spent 77.6% of their time moving forward, while hermaphrodites spent an average of 74.3% of their time moving forward. On average, males spent 15.6% of their time moving backward, and hermaphrodites spent an average of 14.8%. Hermaphrodites spent significantly more time being still than males (p < 0.05), with an average time spent still of 10.8%, while males were, on average, still for 6.8% of the time. n = 140 worms, per sex. Error bars denote standard deviation. (B) Hermaphrodites reverse more often to soft mechanical stimuli than males. (C) Males reverse more often to exposure to high concentrations of butanone than hermaphrodites.
