bir-1, the C. elegans orthologue of vertebrate Survivin is expressed from. an operon together with transcription and splicing cofactor CeSKIP (
skp-1). and is also expressed in postmitotic cells. We have previously shown that. BIR-1 has developmental roles and interacts with vertebrate thyroid. receptor in regulation of gene expression from thyroid hormone responsive. promoters. In this project, we analyzed the proteomic changes in L1 larvae. over-expressing functional
bir-1 from an extra-chromosomal array.. Complete proteomes of synchronized larvae were analyzed using two. dimensional chromatography consisting of chromatographic isoelectric. focusing as the first dimension separation and reversed phase. chromatography as the second dimension separation. Our results show that. over-expression of BIR-1 induces gross proteomic changes despite the lack. of an observable developmental phenotype. We are currently analyzing. protein fractions that show altered chromatographic characteristics in BIR-. 1 over-expressing animals using mass spectroscopy.. Acknowledgement: We thank Drs. A.
