In this issue of Cell, Wu etal. employed C.elegans and human cell experiments to identify a pathway through which metformin increases lifespan and inhibits growth. A key transcriptional target, ACAD10, is activated when metformin induces nuclear exclusion of the GTPase RagC, thereby inhibiting mTORC1 through an unexpected mechanism.

We have previously shown that egl-44 and egl-46 are two transcription regulators involved in the development of several types of neurons in C. elegans, with significant homology in other organisms, and that they repress the expression of touch cell-specific features in FLP neurons (Wu et al., 2001,Genes. The microtubule organizing center, or centrosome, is an unusual organelle. Unlike most organelles, it is not bounded by a membrane, yet it is distinct from the surrounding cytoplasm. It is at the center of important processes in animal and fungal cells, yet many plant cells completely lack it. And perhaps most perplexingly, the centrosome duplicates precisely once per cell cycle, yet the molecular mechanism of duplication remains obscure.

Nematodes are strongly attracted towards E. coli (1). The movement of a population of nematodes from the center of an agar plate towards bacteria at the circumference can be quantitatively analyzed by a sequence of first order rates (2,3), analogous to radioactive decay. Thus, individual nematodes under these conditions do not behave identically. As time goes on, the nematodes remaining at the center will be slower and slower (viz: Dave Hirsh's question, last Newsletter) . Rate constants for leaving the center and the intermediate zone are unique for different mutants. The method may be used as an enrichment technique for paralyzed as well as for chemotaxis-defective mutants.
