In wild type animals we observed distinct AMAN-2::GFP structures in the ASI cells. However, in the majority of the sql-1(tm2409) animals the AMAN-2::GFP signal was more fragmented or even diffuse.

gpa-3QL : sql-1

In sql-1(tm2409) animals, AMAN-2::GFP signal was more fragmented or even diffuse than observed in control animals, very similar effects were seen in sql-1(tm2409); gpa-3QL animals.

Authors examined the expression level of mannosidase II, a glycosidase localized in the medial Golgi stack using pwIs503[vha-6p::mans::gfp] and found it was severely reduced. pwIs503 contains a signal sequence and a Golgi membrane-anchor domain of C. elegans alpha-mannosidase II (aman-2) fused to GFP, driven by the vha-6 promoter.
