POP-1 binds to a fragment of
enh740 in vitro (nucleotide positions -4001 to -6644 with respect to the ATG translation start codon of
ceh-13 in the wild-type sequence. Radio-labeled wt-pop formed a complex with POP-1
(aa180-280) (Fig. 6B, lane 2) This complex could be competed for with unlabeled wt-pop (Fig. 6B, lane 3) but not with mut-pop (Fig. 6B, lane 4), an oligo identical with wt-pop except for a 3-bp substitution in the putative core binding site (Fig. 6A), indicating that POP-1
(aa180-280) binds specifically to this sequence.
