Mus musculus PHENOTYPE: Homozygous KO and poly-alanine KI is embryonic lethal. Heterozygosity affects mitochondria and causes age- and muscle-specific muscle hypo- and hypertrophy. [provided by MGI curators]
- park [Search on AGR]
Drosophila melanogaster parkin (park) encodes an E3 ubiquitin ligase with a key role in protein ubiquitination. It is involved in mitochondrion organization, oxidative stress and locomotion.

Homo sapiens This gene encodes a protein that interacts with the forkhead-associated domain of the Ki-67 antigen. The encoded protein may bind RNA and may play a role in mitosis and cell cycle progression. Multiple pseudogenes exist on chromosomes 5, 10, 12, 15, and 19.[provided by RefSeq, Jan 2009]
- porin [Search on AGR]
Drosophila melanogaster porin (porin) encodes a beta-barrel channel protein localized in the outer membrane of mitochondria that conducts translocation of various ions and metabolites between cytoplasm and mitochondria. It plays roles in mitochondria dynamics and mitophagy, and is ubiquitinated by the E3 ligases encoded by park and Mul1.
- clu [Search on AGR]
Drosophila melanogaster clueless (clu) encodes a cytoplasmic ribonucleoprotein that is required for mitochondrial function. It functions in part with protein complexes at the outer mitochondrial membrane, such as the ribosome, the translocase complex TOM and the mitophagy proteins encoded by park and Pink1. Loss of clu causes mitochondrial oxidative damage, decreased ATP and lethality.

Homo sapiens The small G protein GDP dissociation stimulator (smg GDS) is a regulator protein having two activities on a group of small G proteins including the Rho and Rap1 family members and Ki-Ras; one is to stimulate their GDP/GTP exchange reactions, and the other is to inhibit their interactions with membranes. The protein encoded by this gene contains 9 'Armadillo' repeats and interacts with the smg GDS protein through these repeats. This protein, which is highly concentrated around the endoplasmic reticulum, is phosphorylated by v-src, and this phosphorylation reduces the affinity of the protein for smg GDS. It is thought that this protein serves as a linker between human chromosome-associated polypeptide (HCAP) and KIF3A/B, a kinesin superfamily protein in the nucleus, and that it plays a role in the interaction of chromosomes with an ATPase motor protein. Several transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Mar 2011]
