Homo sapiens Cell motility, adhesion, cytokinesis, and other functions of the cell cortex are mediated by reorganization of the actin cytoskeleton and several formin homology (FH) proteins have been associated with these processes. The protein encoded by this gene contains two FH domains and belongs to a novel FH protein subfamily implicated in cell polarity. A key regulator of cytoskeletal architecture, the small GTPase Rho, is activated during development by Wnt/Fz signaling to control cell polarity and movement. The protein encoded by this gene is thought to function as a scaffolding protein for the Wnt-induced assembly of a disheveled (Dvl)-Rho complex. This protein also promotes the nucleation and elongation of new actin filaments and regulates cell growth through the stabilization of microtubules. Alternative splicing results in multiple transcript variants encoding distinct proteins. [provided by RefSeq, Jul 2012]

Homo sapiens This gene encodes a protein which is a member of the formin/diaphanous family of proteins. The gene is ubiquitously expressed but is found in abundance in the spleen. The encoded protein has sequence homology to diaphanous and formin proteins within the Formin Homology (FH)1 and FH2 domains. It also contains a coiled-coil domain, a collagen-like domain, two nuclear localization signals, and several potential PKC and PKA phosphorylation sites. It is a predominantly cytoplasmic protein and is expressed in a variety of human cell lines. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Dec 2015]

Rattus norvegicus Enables fumarate hydratase activity. Predicted to be involved in several processes, including dicarboxylic acid metabolic process; positive regulation of cold-induced thermogenesis; and positive regulation of double-strand break repair via nonhomologous end joining. Predicted to act upstream of or within homeostasis of number of cells within a tissue. Predicted to be located in cytosol; nucleus; and site of double-strand break. Predicted to be active in mitochondrial matrix. Human ortholog(s) of this gene implicated in fumarase deficiency and lung non-small cell carcinoma. Orthologous to human FH (fumarate hydratase); PARTICIPATES IN altered citric acid cycle pathway; citric acid cycle pathway; fumaric aciduria pathway; INTERACTS WITH (+)-schisandrin B; (R)-adrenaline; 1,2,4-trimethylbenzene.
