spe-46 is localized to the spermatogenic tissue. In gonads from hermaphrodite L4 larvae, GFP is present in a very specific population of cells, likely those that will produce the sperm. In the male gonad, GFP labeling begins at approximately mid to late pachytene stage, and it is present through the appearance of the sperm. Both primary and secondary spermatocytes contain GFP, as do the sperm.

Expression is first seen in this strain in the 50 - 80 cell stage embryo and is not expressed beyond L1 stage larvae. The cells in which expression occurs are as yet unidentified. They mostly occur in pairs or quadruplets. A number of nuclei stain through to the 3-fold stage embryo, but only one staining nucleus is seen in L1 larvae. We will look into this pattern in greater detail.

Strikingly, analysis of the previously uncharacterized H3.3 homologs revealed that his-70 and his-74 show germ line-restricted expression patterns. His-74 expression is restricted to the germ line at all stages of development in both males and hermaphrodites. HIS-74::GFP is only faintly visible in early embryos, and robustly reappears in the primordial germ cells (PGCs; the germ line precursor cells that will give rise to the entire germ line of the worm) and faintly in some cells surrounding the PGCs.

Visible expression of the pst-1 reporter is first seen at around the 2-fold stage. Expression of the pst-1 reporter is detected in the lateral blast cells that will later either fuse to the hypodermal syncytium or give rise to seam cells, a group of specialized hypodermal cells. Expression continues to be detected throughout life in the seam cells and in pharyngeal gland cells. In addition, weaker expression is seen in a number of unidentified cells in the head, possibly in amphid support cells and the duct and/or pore cell, as well as, possibly, in the intestine.

Animals carrying the mir-48::gfp construct showed temporal expression in the hypodermal seam cells two stages earlier than its let-7 homologues at the L2 stage. mir-48 was not detected in the gonad at any stage. specific expression of mir-48 was observed in the P5.p and P7.p cells that will assume 2 vulval cell fates. It is interesting to note that expression of mir-237, mir-84, and mir-48 was absent in the P6.p cell during the mid- to late L3 stage, and expression of mir-237 and mir-84 but not mir-48 was present in the anchor cell.

Furthermore, the spatial patterning of lin-4 in the hypodermal seam cells correlated with the requirement of this gene at the L1/L2 transition to direct normal seam cell development.lin-4 expression was first observed within the developing gonad at the beginning of L3, specifically in the distal tip cells and the anchor cell of the somatic gonad and expression in the distal tip cells continued in the adult. lin-4 and let-7 were weakly expressed in the anchor cell, which is required for proper vulval patterning, and in the P5.p, P6.p, and P7.p cells that will later differentiate into the mature vulva.

To determine whether the T27A3.1pro::gfp neurons were chemosensory amphid and/or phasmid neurons, young adult worms were stained using the lipophilic dye, carbocyanine DiD. Colocalization of DiD staining with GFP was found in the head and the tail. Based on their positions, the chemosensory neurons expressing T27A3.1 are ASK L, ASK R, ASI L, and ASI R. In addition, at least two other T27A3.1-expressing neurons located anterior and posterior to the DiD-filled, ASH L neuron were identified as ADF L and ASE L chemosensory neurons. Other T27A3.1-expressing neurons, and their processes were found alongside DiD-stained chemosensory neurons and will require further analysis to be identified. In the tail region, T27A3.1pro::gfp was found in the more anterior DiD-filled neurons corresponding to PHB L and PHB R phasmid neurons.

ref-2::venus is detected in a substantial number of dividing cells in the embryo. At the end of gastrulation, the reporter is detected in some neuroblasts on the ventral side of the embryo. These neuroblasts include the left/right symmetric pair of ABpl/rpapaaa neuroblasts, which will give rise to AIY and its sister cell, the SMDD motor neuron, through an asymmetric cell division. During interphase, the REF-2::VENUS protein is detected in the nucleus of the SMDD/AIY mothers, then spreads into the cytoplasm just before cleavage. No expression is observed in AIY during larval and adult stages. Thus ref-2 appears to be expressed only transiently in the AIY lineage during embryogenesis. REF-2::VENUS is also expressed in the excretory system (G1, G2, excretory pore, and excretory gland) in the P cell lineage and in the Y and B cells.
