A MEC-19:: GFP translational fusion was found throughout the TRN neurite and also on the plasma membrane and spots within the TRN cell body; its expression overlapped only partially with MEC-4 and MEC-2 (Topalidou and Chalfie 2011) in the proximal neurite and cell body. In the cell body, MEC-19 spots also were found to partially overlap with the Golgi marker AMAN-2::TagRFP.
POML-1 was expressed in only a few neurons. In addition to the six touch receptor neurons (Topalidou and Chalfie, 2011), POML-1 was found in the IL1, AIM, ALN, and BDU neurons. In touch receptor neurons, POML-1 localizes to the endoplasmic reticulum. Rescuing and tagged translational fusions of POML-1 are primarily expressed in the TRN cell body, with weak diffuse expression and puncta in the proximal TRN neurite. Consistent with the ER localization of MEC-6, the protein was absent from the TRN surface.
In the meiotic cells of the gonad including oocytes and in the interphase cells of 200-cell embryos, DNA topoisomerase I was localized in nuclei. At the comma stage, a more narrowed concentration of DNA topoisomerase I was observed in the nucleoli of gut cells.
GFP was shown to express in the excretory cells, coelomocytes, head neurons, hypodermal cells, and in the pharynx of C. elegans. This expression pattern was further confirmed by antibody staining using polyclonal antibodies against UCH/CeUBP130. Excretory cells were identified by the 'I-i' shape GFP expression in the head region). GFP was also shown to express in coelomocytes.
