pal-1 RNA evenly distributed in 1- and 2-cell embryos. In 4-cell embryos, pal-1 RNA was either uniformly distributed (approximately 50%) or preferentially localized to the posterior blastomere. Nuclear localized transcripts were not detected before the 24-cell stage. In 6 cell and older embryo, pal-1 RNA was more abundant in cells that express PAL-1 protein.

We detected embryonic expression of our rescuing pal-1::gfp construct, containing the full genomic sequence of pal-1, in two pairs of epidermal daughter cells, V3 and P7/8, and V4 and V6, from shortly after their birth. In contrast, we were unable to detect any expression of pal-1 in L1 larvae, supporting the hypothesis that pal-1 functions in embryos to coordinate L1 ventral epithelial morphogenesis.

DRAG-1::GFP was present in pharyngeal, hypodermal and intestinal cells. Robust expression in the M lineage was also detected from the 1-M to 4-M stage. DRAG-1::GFP signal was reduced (40%, n=20) or undetectable (60%, n=20) at the 8-M stage, and completely undetectable beyond the 8-M stage.

UNC-130::GFP expression is not detectable in the early M lineage from the 1-M to the 4-M stage. Beginning at the 8-M stage and continuing through the 18-M stage, UNC-130::GFP expression is visible in all the ventral, but not in any of the dorsal, M lineage cells. It was noticed that M.vlp and M.vrp and their descendants exhibited relatively higher levels of UNC-130::GFP expression than their anterior counterparts.
