The Pmir-83::GFP reporter was widely expressed at all stages of development and across a range of tissues such as the intestine, neurons, and body wall muscles. Furthermore, we observed a significant decrease in the expression of miR- 83 between the L3 and L4 stages, which is consistent with a previous report (Martinez et al., 2008a,b), and a further decrease from day 1 of the adult stage through day 12. About 44% decrease in GFP expression driven by miR-83 was observed in older adults (day 12) compared to young adults (day 4).
This construct was expressed faintly in QL and QR, more strongly in the neighboring epidermal cells (dorsal hyp7 cells, ventral P cells and lateral V cells), and in dorsal and ventral body muscle cells.
Staining was observed in all larval and adult stages, localizing to the nuclei of the hypodermal cells, specifically the head (hyp3, hyp4, hyp6, and hyp7) and seam/body (hyp7, H, V, and P) hypodermal cells.
