vang-1::YFP reporter is expressed in the VPC progeny. Bright vang-1::YFP-expressing cell was also observed in ventral cord neuron.

vang-1 is expressed in VNC neurons. VANG-1 (n = 9) was observed to localize to cell membranes and is enriched at contracting cell contacts. In addition, VANG-1 is localized at rosette foci. These localization dynamics resemble that of endogenous myosin II (NMY-2) (Dickinson et al., 2013), which is also found on contracting cell contacts and rosette foci. VANG-1 exhibited approximately 4-fold enrichment at contracting edges versus non-contracting edges and approximately 7-fold enrichment at the rosette center relative to the planar contacts between cells engaged in the rosette.

vang-1 transcripts can be detected in both the QL and QR neuroblasts at the onset of larval development. During the initial polarization and migration of the Q neuroblasts that precedes their response to Wnt ligands [35, 36], there is a clear increase in vang-1 expression in both neuroblasts to an average of 4 ± 1.5 transcripts. After division, there is a significant decrease in vang-1 expression in QL.p (1.5 ± 1 transcripts) and QR.p (1.5 ± 1 transcripts), but this expression remains relatively constant during QR.p and QR.pa migration.

In L4 larvae, vang-1 promoter activity was detected in many head and ventral cord neurons, some bilaterally located mid-body neurons including the HSNs, as well as non-neuronal tissue such as somatic gonad cells, uterine cells, vulval muscle and vulval epithelial cells. Co-expression with the VC1-6 specific reporter Plin-11::RFP revealed that the subset of ventral cord neurons expressing vang-1 includes all six VC neurons.

sax-3 is expressed in VNC neurons. A SAX-3::GFP transgene showed low levels of expression and GFP signal, making it difficult to quantify. However, in a small fraction of embryos with relatively high signal (n = 2), the localization patterns resemble that of VANG-1 qualitatively (See Expr13290).
