These animals exhibited specific GFP expression in two cells in the head, the position and morphology of which suggested that they were the XXX cells . Colocalization of GFP and RFP in animals coexpressing eak-3::GFP and sdf-9::RFP, which marks the cytoplasm of the XXX cells, confirmed that the eak-3 promoter is specifically active in the XXX cells.

Both in situ hybridization and reporter transgene fusions revealed that end-3 is expressed in the early E lineage. The end-3 reporter is not expressed in maternal mutants in which E fate is not specified and is ectopically expressed in the predicted cells in mutants in which endoderm is made ectopically; in all cases, end-3 expression marks cells that are specified to produce endoderm.

We also identified a striking expression pattern for the helix-loop-helix TF hlh-15, in which expression is confined to a single neuron in the tail (DVA) and a single paired neuron in the head (AVKL/R). A previous study generated a promoter::GFP transgene for hlh-15 and likewise reported expression in a single tail neuron and a single paired head neuron (Grove et al. 2009) but tentatively assigned the expression to the DVA and RIF neurons based on cell position and morphology. To clarify the discrepancy, we obtained the hlh- 15p::GFP transgene and crossed it with nuclear red fluorescent transgenes (H2B::mCherry) expressed in only the DVA neuron or only the AVK neuron (Wang et al. 2017). We found that the DVA mCherry marks the nucleus of the cell identified in the tail, and the AVK mCherry marks the nucleus of the paired head neurons, confirming VISTA's cell-type identification for hlh-15 gene expression.

To determine whether the APC might function at GABA synapses, we analyzed the subcellular localization of a GFP-tagged APC subunit EMB-27 CDC16 (GFP::EMB-27) relative to mCherry-tagged SNB-1 Synaptobrevin (mCherry::SNB-1), which marks presynaptic sites, in GABA neurons (Fig. 7A). We found that approximately 90% of the GFP::EMB-27 puncta co-localized with mCherry::SNB-1 puncta (90.28 ± 14.11%, n = 20 animals) and vice versa (90.11 ± 15.67%, n = 20 animals).

pcdr-1p::GFP expression was observed in ciliated neurons in the head and neurons in the ventral nerve cord (VNC). In the VNC pcdr-1p::GFP expression co-localised with acr-2p::mCherry, which marks the cholinergic motor neurons, however we also observed GFP positive cell bodies which were not colocalised with this marker suggesting that PCDR-1 is expressed in both cholinergic andnon-cholinergic neurons in the VNC. we also observed expression of pcdr-1p::GFP in the vulval muscles and in non-neuronal cells in the tail. To determine the identity of these non-neuronal cells we co-expressed mCherry from a 1.3Kb fragment of the egl-5 promoter that drives expression in rectal epithelial cells. pcdr-1p::GFP expression co-localised with egl-5p::mCherry indicating that PCDR-1 was expressed in the rectal epithelium.
